I then selected "Healer" from the Ocean characters to copy with Theseus, which requires paying another coin. I attempted to use the newly acquired +1 coin purple manta to activate "Healer", but the interface would not allow it.

If raising the last depth of a location (by using that resource) results in gaining a purple wild Manta, the Manta arrives ready to use, and should be able to be used during that same turn, immediately. I have never had a problem with Theseus or Healer, but the Theseus -> Healer sequence might be bugged.
